Kailera cleared its End-of-Phase 2 FDA meeting for oral ribupatide, putting a global Phase 3 obesity program on the calendar for the first half of 2027. R&D spending tripled to $84.7 million as all four candidates stay on track, and management says $1.04 billion in cash funds operations into mid-2028, past the key readouts.

Kailera Therapeutics ended the third quarter of 2026 with no product revenue, as expected for a clinical-stage biotech. The company completed its End-of-Phase 2 meeting with the FDA for oral ribupatide, the clearest milestone yet on the path to a global Phase 3 obesity trial now targeted for the first half of 2027.

The cost of that progress was substantial. Research and development expenses surged to $84.7 million in the quarter, reflecting clinical trial activity across all four GLP-1-based candidates, contract manufacturing commitments, and an expanding workforce. General and administrative expenses also climbed, pushing the net loss for the quarter higher.

The injectable version of ribupatide is already fully enrolled in both a U.S. Phase 2b high-dose trial and the global KaiNETIC Phase 3 program, with data expected in mid-2027 and mid-2028 respectively. Safiglipron's global Phase 2 trial, also fully enrolled, now targets a data readout in mid-2027, and the KAI-4729 tri-agonist is expected to enter a Phase 1 trial before the end of 2026.

Management's confidence rests heavily on the balance sheet. Cash, cash equivalents, and marketable securities stood at $1.04 billion, which the company says is enough to fund operations into mid-2028, a runway that covers the most critical anticipated data readouts across the ribupatide franchise. CEO Ron Renaud stated that all four product candidates are on track to deliver important clinical milestones in 2027, including the initiation of the oral ribupatide Phase 3 trial and high-dose injection Phase 2b data.

For investors, the key variables to monitor are the Phase 3 start date for oral ribupatide in early 2027 and whether the high-dose injection data arriving mid-year supports the differentiation story Kailera is building around the ribupatide franchise. The company operates in a crowded GLP-1 obesity market, and its own filings flag competition, reliance on third-party manufacturers, and the need for additional financing beyond mid-2028 as material risks.
