A pro forma cash pile of roughly $3.4 billion funds Vaxcyte all the way to a potential US launch of VAX-31, which just met every primary endpoint in the pivotal OPUS-1 Phase 3. The loss narrowed against expectations even as R&D climbed on manufacturing scale. FDA scrutiny of serotypes 3 and 12F remains the open question.

Vaxcyte reported a GAAP loss per share of $1.99 for the third quarter of 2026, beating expectations. The net loss widened year over year as research and development spending surged on manufacturing scale-up costs and accelerating OPUS trial activity.

The quarter's headline event was the OPUS-1 pivotal Phase 3 readout for VAX-31, the company's 31-valent pneumococcal conjugate vaccine candidate. In a study of adults aged 50 and older, VAX-31 met all prespecified co-primary immunogenicity endpoints, including noninferiority, superiority, and immunobridging criteria, with a safety profile described as similar to existing vaccines PCV20 and PCV21. CEO Grant Pickering called the results a significant milestone and said they support VAX-31's potential best-in-class profile.

One regulatory wrinkle investors will be tracking: for two serotypes, 3 and 12F, VAX-31 missed the stricter lower bound of the confidence interval criterion of greater than 0.667 when compared against PCV21, though it did clear the historical threshold of greater than 0.5. The FDA's scrutiny of those two serotypes is expected to be a focal point in the regulatory review, making the agency's eventual stance on the comparison a key variable in VAX-31's approval path.

Vaxcyte entered the quarter with a pro forma cash position of roughly $3.4 billion, bolstered by approximately $1.15 billion raised through concurrent October 2026 offerings of common stock, pre-funded warrants, and 1.50% convertible senior notes due 2032. Management has stated this positions the company through a potential U.S. commercial launch of VAX-31 for adults, which matters given the company generates no revenue and faces steadily rising operating costs.

The pipeline calendar ahead is dense. Results from OPUS-2, which studied VAX-31 alongside a flu vaccine, and OPUS-3, which enrolled previously vaccinated adults, are both targeted for the first half of 2027. Infant Phase 2 data for VAX-31 and topline Phase 1 results for VAX-A1, the company's Group A Streptococcus candidate, are also expected by the end of the first half of 2027 and the second half of 2027, respectively. A BLA submission for VAX-31 is planned for the first half of 2028.
