Xencor Q3 2026: One-Time Payments Flip the Company to Profit
As seen on the 24/7 Wall St. homepage on November 4, 2026.
The first profitable quarter in years came courtesy of one-time cash: a $52.5 million Alexion settlement payment plus a $25 million Astellas milestone, with another $52.5 million from Alexion due in Q3 2027. That funds the pipeline into 2029, and management now expects XmAb819 to enter a registration-enabling study in advanced kidney cancer by the end of 2027.

Xencor swung to a GAAP profit of $0.50 per share in Q3 2026, beating a consensus estimate of negative $0.34. Revenue of $116.52 million came in well above what analysts had expected. The quarter marked the company's first profitable period in years.
The headline numbers were driven by a cluster of one-time receipts. A $52.5 million payment under a settlement agreement with Alexion landed in August 2026, and a $25 million milestone from Astellas was triggered when the first patient was dosed in the Phase 3 ASP2138 trial. A smaller $2.5 million milestone from Zenas, tied to a Japanese marketing application for obexelimab, also contributed.
The cash haul has real staying power for the pipeline. Management now expects to end 2026 with between $480 million and $500 million in cash, cash equivalents, and marketable debt securities, with that runway extending into 2029. A final $52.5 million Alexion payment is still owed in Q3 2027, providing another near-term anchor.

The bigger strategic story centers on XmAb819, Xencor's ENPP3 x CD3 bispecific antibody being studied in advanced clear cell renal cell carcinoma. CEO Bassil Dahiyat said the company anticipates XmAb819 will enter a potential registration-enabling study by the end of 2027, following xaluritamig and ASP2138 as the third XmAb 2+1 bispecific to reach that stage. FDA engagement on a potential accelerated approval pathway is planned for the first half of 2027.
Beyond kidney cancer, Xencor's pipeline spans autoimmune and oncology programs. Early dose-escalation data for plamotamab, a CD20 x CD3 T-cell engager being evaluated in rheumatoid arthritis, was set to be presented at ACR Convergence on November 9, 2026. The TL1A program XmAb942 is enrolling in a Phase 2b ulcerative colitis study, with a primary endpoint analysis expected in the second half of 2027.
